My research focuses on the epigenetic basis of cancer, stem cells and human development, especially about how cellular signaling regulates cell growth and differentiation, how the genetic material is elegantly organized in human cells, how gene activities are controlled, and how these events become abnormal in cancer and other diseases. We have been using cutting-edge molecular biology techniques and characterizing genetically modified mouse models to investigate these important issues. In addition, we have been also interested in generating different stem cells by somatic cell reprogramming. The research has provided, and will continue to yield, new insights into how to develop novel means for prevention, diagnosis and treatment of related human diseases.

Our laboratory focuses on the molecular and epigenetic basis of cancer, stem cells and animal development, especially about how cell signaling regulates chromatin modification, gene expression, and other events in the nucleus. His research program is mainly divided into the following three parts:
- Caractérisation de deux histones acétyltransférases paralogues Le gène humain pour MOZ (monocytic leukemia zinc finger protein) a été identifié en 1996 comme un partenaire de fusion réarrangé dans t(8;16)(p11;p13), une translocation récurrente associée aux sous-types M4/M5 de la leucémie aiguë myéloïde. Depuis lors, ce gène a été trouvé comme partenaire dans trois autres translocations chromosomiques liées à la leucémie aiguë myéloïde et aux syndromes myélodysplasiques liés à la thérapie.
En 1999, notre groupe a identifié MORF (MOZ-related factor) comme un paraglogue de MOZ et découvert que MOZ et MORF sont des histones acétyltransférases. Comme le gène MOZ dans t(8;16), le gène MORF est réarrangé chez les patients atteints de leucémie avec t(10;16)(q22;p13). Ces deux gènes sont importants pour différents programmes de développement chez les mammifères tels que le maintien des cellules souches hématopoïétiques et la neurogenèse. MOZ et MORF sont des sous-unités catalytiques de complexes multiprotéiques contenant ING5 (inhibitor of growth 5), EAF6 (homologue de Esa1-associated factor 6), et BRPF1/2/3 (bromodomain-PHD finger protein 1, -2, ou -3). Importamment, ces complexes sont également formés par au moins une protéine de fusion issue d'une translocation chromosomique associée à la leucémie. Le groupe du Dr. Yang a récemment analysé comment l'interaction des sous-unités régule l'activité enzymatique des protéines MOZ et MORF normales et leucémiques.
Characterization of two paralogous histone acetyltransferases The gene for human MOZ (monocytic leukemia zinc finger protein) was identified in 1996 as a fusion partner rearranged in t(8;16)(p11;p13), a recurrent translocation associated with the M4/M5 subtypes of acute myeloid leukemia. Since then, this gene has been found to be a partner in three other chromosomal translocations linked to acute myeloid leukemia and therapy-related myelodysplastic syndromes.
In 1999, our group identified MORF (MOZ-related factor) as a MOZ paraglog and discovered that MOZ and MORF are histone acetyltransfrerases. Like the MOZ gene in t(8;16), the MORF gene is rearranged in leukemia patients with t(10;16)(q22;p13). These two genes are important for different developmental programs in mammals such as maintenance of hematopoietic stem cells and neurogenesis. MOZ and MORF are catalytic subunits of multiprotein complexes containing ING5 (inhibitor of growth 5), EAF6 (homolog of Esa1- associated factor 6), and BRPF1/2/3 (bromodomain-PHD finger protein 1, -2, or -3). Importantly, these complexes are also formed by at least by one fusion protein from a leukemia-associated chromosome translocation. Dr. Yang’s group has recently analyzed how subunit interaction regulates the enzymatic activity of normal and leukemic MOZ and MORF proteins.
- Fonction et régulation des histones désacétylases Les histones désacétylases (HDACs) sont des enzymes relativement nouvelles capables d'éliminer les groupes acétyle des formes acétylées des histones et de nombreux autres substrats protéiques. Dans le système mammifère, dix-huit protéines ont été démontrées posséder une telle activité enzymatique, dont quatre d'entre elles (HDAC4, 5, 7 et 9) forment un sous-groupe connu sous le nom de classe IIa.
En 1999, notre groupe a identifié HDAC4 et a montré que cette désacétylase interagit avec la famille de régulateurs transcriptionnels MEF2 (myocyte enhancer factor 2) pour réprimer la transcription (le processus de production d'ARN messager à partir des gènes). Cette répression est étroitement contrôlée dans les cellules : les kinases dépendantes du signal phosphorylent les HDACs de classe IIa, entraînant ainsi l'exportation nucléaire de ces enzymes et favorisant la transcription dépendante de MEF2. Cette interaction dynamique est essentielle dans différents tissus, car la famille MEF2 joue un rôle crucial dans un certain nombre de programmes cellulaires, notamment la différenciation des cellules musculaires et osseuses, la survie neuronale, la mort programmée des cellules T et l'intégrité endothéliale. Il existe quatre isoformes de MEF2 chez les humains, MEF2A, B, C et D. Le groupe du Dr. Yang étudie comment la signalisation cellulaire contrôle l'action de HDAC4 et de ses paralogues en termes de régulation de MEF2.
Function and regulation of histone deacetylases Histone deacetylases (HDACs) are relatively novel enzymes that are able to remove acetyl groups from acetylated forms of histones and many other protein substrates. In the mammalian system, eighteen proteins have been shown to possess such enzymatic activity, with four of them (HDAC4, 5, 7 and 9) forming a subgroup known as class IIa.
In 1999, our group identified HDAC4 and showed that this deacetylase interacts with the MEF2 (myocyte enhancer factor 2) family of transcriptional regulators to repress transcription (the process of making messenger RNA from genes). This repression is tightly controlled in cells: signal-dependent protein kinases phosphorylate class IIa HDACs, consequently driving nuclear export of these enzymes, and promoting MEF2- dependent transcription. This dynamic interplay is essential in different tissues as the MEF2 family plays a crucial role in a number of cellular programs, including muscle and bone cell differentiation, neuronal survival, programmed T cell death, and endothelial integrity. There are four MEF2 isoforms in humans, MEF2A, B, C and D. Dr. Yang’s group has been investigating how cell signaling controls the action of HDAC4 and its paralogs in terms of MEF2 regulation.
- Modification post-traductionnelle dans la différenciation cellulaire et la reprogrammation Par analogie avec de nombreuses modifications présentes dans les histones et le suppresseur de tumeurs p53, nous avons récemment postulé que cela se produit également chez d'autres régulateurs protéiques majeurs. En utilisant MEF2 comme exemple, notre groupe a identifié un motif de sumoylation dépendant de la phosphorylation conservé dans la protéine MEF2 du ver à l'homme. Un motif similaire est présent dans les facteurs de reprogrammation des cellules souches KLF4 et Sox2. Ainsi, le groupe du Dr. Yang évalue l'importance d'un tel motif dans la reprogrammation des fibroblastes en cellules souches pluripotentes induites (iPS). Post-translational modification in cell differentiation and reprogramming By analogy to numerous modifications present in histones and the tumour suppressor p53, we have recently postulated that this also occurs in other major protein regulators. By use of MEF2 as an example, his group found a phosphorylation-dependent sumoylation motif conserved in MEF2 protein from worms to humans. A similar motif is present in the stem cell reprogramming factors of KLF4 and Sox2, so Dr. Yang’s group has been assessing the importance of such a motif in reprogramming fibroblasts into induced pluripotent stem (iPS) cells
