working on
Our lab addresses fundamental questions in cell biology to understand mechanisms of breast cancer initiation and progression. His lab utilizes animal models, 3D organoids, and patient-derived tissue to interrogate dynamic behaviors between normal or cancer cells and their microenvironment. A goal of this research is to understand how cell and tissue organization influence the growth and invasive properties in cancer, to improve predictive, preventative, and therapeutic options.
- Caractérisation de l'Hétérogénéité Cellulaire dans la Progression Précoce du Cancer du Sein Le carcinome canalaire in situ (CCIS) est un précurseur non obligatoire du cancer invasif. Cela signifie que le cancer invasif se développe à partir du CCIS, mais que tous les CCIS ne progresseront pas vers un cancer invasif. Il n'existe actuellement aucune caractéristique clinique, pathologique ou génétique établie permettant de prédire de manière fiable la progression du CCIS vers un cancer invasif. Characterizing Cell Heterogeneity in Early Breast Cancer Progression Ductal Carcinoma In Situ (DCIS) is a non-obligate precursor for invasive cancer. This means that invasive cancer develops from DCIS, but not all DCIS will progress to invasive cancer. There are currently no established clinical, pathological, or genetic features that reliably predict progression of DCIS to invasive cancer. We envision that epithelial phenotypic diversity and plasticity contribute to progression of DCIS to invasive cancer by endowing cells the ability to adapt from a ductal environment to a stromal environment. The major objective of this theme is to elucidate the molecular mechanisms underlying early breast cancer development with the long-term goal of developing predictive biomarkers for DCIS
- Signalisation de la Polarité Cellulaire et Transformation Maligne La perte de l'organisation tissulaire et de la polarité apico-basale (c'est-à-dire la ségrégation des domaines membranaires apicaux et basolatéraux) sont des caractéristiques des cancers épithéliaux. Un concept que nous avons promu est que la perte de la polarité apico-basale dérègle les voies de signalisation impliquées dans l'homéostasie épithéliale, ce qui constitue une étape permissive dans la progression vers un état malin. La polarité membranaire apico-basale délimite des compartiments membranaires cellulaires distincts, avec une membrane apicale qui fait face à la lumière et une membrane basolatérale qui est en contact avec les cellules adjacentes et la matrice extracellulaire. Les régulateurs clés de la polarité apico-basale contrôlent un large éventail d'activités cellulaires, notamment la prolifération, la survie, la motilité et l'invasion, qui sont importantes pour la progression du cancer du sein. Cell Polarity Signaling and Malignant Transformation Loss of tissue organization and apical-basal polarity (i.e. the segregation of apical and basolateral membrane domains) are characteristics of epithelial cancers. A concept that we have championed is that loss of apical-basal polarity deregulates signaling pathways involved in epithelial homeostasis, which is a permissive step in the progression to a malignant state. Apical-basal membrane polarity demarcates distinct cellular membrane compartments, with an apical membrane that faces the lumen and a basolateral membrane that contacts adjacent cells and the extracellular matrix. Key regulators of apical-basal polarity control a wide range of cellular activities, including proliferation, survival, motility, and invasion that are important for breast cancer progression. This theme investigates novel mechanisms by which tissue organization and polarity are regulated during cancer progression
- Surmonter les Microenvironnements Suppressifs dans la Progression du Cancer du Sein Le carcinome canalaire in situ (CCIS) se caractérise par un microenvironnement épithélial et immunitaire suppressif qui restreint l'invasion cellulaire. Les macrophages peuvent se trouver dans plusieurs états avec des activités diverses, notamment l'inflammation, la régulation immunitaire, l'angiogenèse, l'invasion et la métastase. Un nombre accru de macrophages associés à la tumeur est associé à un mauvais pronostic chez les patientes atteintes d'un cancer du sein. Les conduits mammaires sont organisés sous forme de deux couches de cellules épithéliales luminales entourées d'une couche myoépithéliale contractile. Dans la progression précoce du cancer du sein, les cellules luminales acquièrent des propriétés malignes et se multiplient. On suppose que les cellules myoépithéliales sont des répresseurs naturels de la progression tumorale en agissant comme une barrière physique pour empêcher les cellules luminales de s'envahir dans les tissus environnants. La perte de cellules myoépithéliales est une caractéristique du passage du CCIS au cancer invasif, cependant, les mécanismes moléculaires qui perturbent la couche myoépithéliale restent largement inconnus. Overcoming Suppressive Microenvironments in Breast Cancer Progression Ductal carcinoma in situ (DCIS) is characterized by a suppressive epithelial and and immune microenvironment that restricts cell invasion. Macrophages can exist in multiple states with diverse activities, including inflammation, immune regulation, angiogenesis, invasion, and metastasis. Increased numbers of tumor-associated macrophages are associated with poor outcome in breast cancer patients. Breast ducts are organized as a bilayer of luminal epithelial cells surrounded by a contractile myoepithelial layer. In early breast cancer progression, luminal cells acquire malignant properties and expand in number. Myoepithelial cells are hypothesized to be natural repressors of tumour progression by acting as a physical barrier to prevent luminal cells from invading into surrounding tissue. Loss of myoepithelial cells is a characteristic of the transition from DCIS to invasive cancer, however, the molecular mechanisms that disrupt the myoepithelial layer remain largely unknown. This theme investigates the mechanisms by which the suppressive immune microenvironment and myoepithelial cells are reprogrammed to permit invasive breast cancer
